เกม Duke Nukem Forever เป็นเกมซูตติ้งมุมมองบุคคลที่ 1 กล่าวถึง โลกกำลังถูกคุกคามโดยเหล่าเอเลี่ยนอีกครั้ง ทำให้ Duke Nukem ตัวละครเอกกล้ามโตของเรื่อง เดินทางกำจัดและทำลายล้างตั้งแต่พวกเอเลี่ยนตัวธรรมดาจนถึงบุกเข้ารังเอเลี่ยน ที่กบดานอยู่ชั้นพื้นโลก นอกจากนี้ยังมีหน่วยจู่โจมและเหล่าสาวๆ หน้าอกโตคอยให้สนับสนุน และให้กำลังใจแบบแนบเนื้อด้วย ทีมผู้สร้างให้ข้อมูลว่าเกมนี้ใช้ทุนประมาณ 20-30 ล้านดอลลาร์สหรัฐ (ประมาณ 600 ล้านบาท - 900 ล้านบาท) โดยนับรวมถึงงบประมาณที่สิ้นเปลืองไปจากการเปลี่ยนแปลงทีมพัฒนา และเปลี่ยนเอนจิ้นซ้ำแล้วซ้ำอีกด้วย
