Assassin's Creed Syndicate กล่าวถึงเมืองลอนดอนในปี 1868 ยุคปฏิวัติด้านอุตสาหกรรม ที่มีสองพี่น้องฝาแฝดชื่อว่า Jacob Frye และ Evie Frye ได้ตั้งแก๊งอาชญากรของตัวเองขึ้นมาเพื่อโค่นล้มอำนาจการปกครองของเทมพลาร์เพื่อปลดปล่อยอิสระภาพให้แก่ชาวลอนดอนที่ถูกกดขี่ ผู้เล่นจะได้เล่นทั้ง Jacob และ Evie ซึ่งสามารถสลับตัวละครไปมาระหว่างเล่นได้ มี Skill Tree แยกเป็นของตัวเอง และทีมพัฒนายังนำเสนอระบบยานพหานะแบบใหม่ที่ผู้เล่นสามารถขโมยได้แบบเกม Grand Theft Auto รวมไปถึงการใช้สายสลิงที่ทำให้ผู้เล่นปีนขึ้นยอดตึกได้เร็วขึ้น
