Assassin's Creed: Syndicate กล่าวถึงเมืองลอนดอนในปี 1868 ยุคปฏิวัติด้านอุตสาหกรรม ที่มีสองพี่น้องฝาแฝดชื่อว่า Jacob Frye และ Evie Frye ได้ตั้งแก๊งอาชญากรของตัวเองขึ้นมาเพื่อโค่นล้มอำนาจการปกครองของเทมพลาร์เพื่อปลดปล่อยอิสระภาพให้แก่ชาวลอนดอนที่ถูกกดขี่ ผู้เล่นจะได้เล่นทั้ง Jacob และ Evie ซึ่งสามารถสลับตัวละครไปมาระหว่างเล่นได้ มี Skill Tree แยกเป็นของตัวเอง และทีมพัฒนายังนำเสนอระบบยานพหานะแบบใหม่ที่ผู้เล่นสามารถขโมยได้แบบเกม Grand Theft Auto รวมไปถึงการใช้สายสลิงที่ทำให้ผู้เล่นปีนขึ้นยอดตึกได้เร็วขึ้น

Assassin's Creed: Syndicate - Jack the Ripper เนื้อเรื่องได้พูดถึงเหตุการณ์หลังจาก Assassin’s Creed Syndicate ประมาณ 20 ปีเมืองหลวงที่สวยงามและเงียบสงบอย่างกรุงลอนดอลประเทศอังกฤษได้มีเหตุการณ์ที่น่ากลัวเกิดขึ้นเมื่ออยู่ๆมีคดีปริศนาที่ฆาตกรนามว่า "Jack the Ripper"ได้ออกไล่สังหารผู้คนซึ่งจะเน้นเป็นผู้หญิงซะส่วนใหญ่ เรียกได้ว่าเป็นคดีที่ทำเอาผู้คนที่อยู่ในช่วงเวลานั้นต่างหวาดกลัวงานนี้ผู้เล่นเลยจะต้องสวมบทเข้าร่วมกองกำลัง FrederickAbberline แห่ง Scotland Yard เพื่อออกตามล่า Jack
