Deus Ex: Human Revolution Director's Cut ได้เพิ่มภารกิจ Tongss Rescue และ Missing Link รวมไว้ในเกมแพคเกตเดียว อีกทั้งทางทีมพัฒนายังปรับปรุงในเรื่องความสมดุลของเกมหลายๆอย่าง ไม่ว่าจะเป็นการต่อสู้กับบอส ,เพิ่มความฉลาดของ AI ,ปรับปรุงกราฟฟิคให้ดูดีขึ้น เนื้อเรื่องย่อจากภาคหลัก ผู้เล่นจะได้รับบทเป็น Adam Jensen ซึ่งอยู่หน่วยรักษาความปลอดภัยของบริษัทพัฒนาเทคโนโลยีชีวภาพแห่งหนึ่ง ซึ่ง Adam Jensen ได้ไปเห็นความลับสำคัญของบริษัท จึงได้กลายเป็นพยานสำคัญที่จะเปิดเผยแผนการอันเลยร้ายของผู้ไม่ประสงค์ดีต่อโลกพระเอกของเราจึงต้องต่อสู้เพื่อเอาชีวิตรอดจากการถูกตามล่าและเพื่อเปิดเผยความจริงให้ชาวโลกได้รับรู้
