เมื่อเหล่าปีศาจทั้งหลายเข้าครอบครองสถานีวิจัย Union Aerospace Corporation (UAC) บนดาวอังคาร คำสาป และความสยองได้เริ่มต้นขึ้น และคุณคือหนึ่งในผู้รอดชีวิตที่ยังเหลืออยู่ ภายใต้ความหวาดกลัวและน่าขนลุก คุณต้องสู้ และสู้ ! เพื่อกำจัดพวกมันให้หมดไป
Doom 3 ยังคงเป็นเกมแนว 3D First Person Shooting ทั่วๆ ไป แต่ความเป็นแอ็คชั่น 3 มิติแทนมุมมองสายตาของผู้เล่นอันเป็นเอกลักษณ์ของ Doom นั้นได้ผสมเข้ากับความรู้สึกที่ตื่นเต้นสยองขวัญชนิดเขย่าประสาทผู้เล่นในขณะที่เล่นตามแบบฉบับของเกมในแนว Horror Adventure ได้เป็นอย่างดี
