ตัวละครที่ผู้เล่นจะได้เล่นใน Assassins Creed Revelations ภาคนี้จะเป็นการรวมตัวนักฆ่าจากทุกๆภาคเอาไว้ นั่นก็คือ Altair จากภาคแรก Ezio จากภาคสอง และ Desmond Miles จากยุคปัจจุบัน เนื้อเรื่องของภาคนี้จะเป็นการรวมกันของทุกๆภาค โดยเริ่มจากช่วงแรก ต้องเล่นเป็น Altair หลังจากนั้นบทจะตัดมาให้เล่นเป็น Ezio ในช่วงเนื้อเรื่องหลังภาค 2 และภาค Brotherhood นั่นคือหลังจบศึกกับทหารกรุงโรม แล้ว คราวนี้ Ezio ต้องต่อสู้กับ Constantinople แห่งจักรวรรดิ์ Ottoman Empire และสุดท้ายจะเป็นเนื้อเรื่องของ Desmond ในยุคปัจจุบัน ซึ่งเป็นเรื่องราวต่อจากภาค Brotherhood เช่นกัน สำหรับใครที่เป็นแฟนตัวยงเกม Assassin's Creed และอยากรู้เนื้อเรื่องช่วงต่อหลังจากตอนจบในภาคเก่าๆล่ะก็ห้ามพลาด Assassins Creed Revelations เด็ดขาด
