Terminator: Resistance Annihilation Line จะเป็นเรื่องราวของ Kyle Reese ตัวละครเอกจากภาพยนตร์ Terminator ภาคแรก โดยเหตุการณ์จะเกิดขึ้นในช่วงกลางของเนื้อหาหลักที่หลังจากกลุ่มผู้ต่อต้านสามารถปราบผู้ที่แผงตัวเข้ามาได้สำเร็จ และ John Conner ก็มีคำสั่งโดยตรงมาให้ทีมของ Jacob River ไปยังฐานที่มั่นใน Northridge หลังจากพบว่าประชาชนที่อยู่ในนั้นหายตัวไปอย่างลึกลับ ซึ่งเขาต้องร่วมมือกับ Reese ผู้เป็นมือขวาของ John ในการเข้าไปสืบหาสาเหตุว่าเกิดอะไรขึ้นในฐานที่มั่นแห่งนี้
