Ezio Auditore da Firenze ชายหนุ่มอายุ 17 ปี ต้องต้องหลบหนีการตามล่าหลังจากที่พ่อและน้องของเขาถูกสังหาร หนำซ้ำครอบครัวของก็เขาถูกทรยศ จากนั้นก็ได้รู้ว่าพ่อของเขาไม่ใช่แค่นายธนาคารธรรมดา ที่จริงแล้วเขาคือหนึ่งในสมาชิกขององค์กรนักฆ่า หลังจากที่ได้รับการช่วยเหลือจากอาของเขา Ezio จึงได้ตัดสินใจเป็นนักฆ่าเต็มตัวเพื่อแก้แค้น และไขปริศนาต่างๆ ที่พ่อของเขาได้ทิ้งไว้
