ในภาคนี้ได้มีการเพิ่มภารกิจต่างๆ ทำให้เกมส์นี้มีความสนุกมากยิ่งขึ้น พร้อมทั้งเพิ่มอิสระในการเล่นที่มากยิ่งขึ้นด้วย ตัวเอกของเราสามารถซ่อนตัวใต้น้ำหรือแม้กระทั่งลากศัตรูลงน้ำก่อนแล้วค่อยเชือด, Ezio สามารถเรียบรู้การป้องกันตัวเองตัวย กำปั้น หรือ หมัด แต่คงไม่ใช่แบบมวยไทย นะครับ เพราะว่าคุณต้องเล่นเป็นนักฆ่าที่สามารถป้องกันตัวเองได้ทุกรูปแบบและ สถานการณ์, ในภาคนี้ได้มาการเพิ่มยานพาหนะขึ้นนั้นคือ เครื่องร่อนนั้นเอง ซึ่งทำให้คุณสามารถเข้าไปในที่ๆ ไม่สามารถเดินเท้า หรือ ปีนป้ายเข้าไปได้ ทั้งนี้คุณยังสามารถกระโดดจากที่สูง เหมือนในภาคแรก แต่ในภาคสองนี้คุณสามารถดิ่งลงน้ำได้อีกด้วย
