กำเนิดระเบิดมือนั้นไม่ปรากฏหลักฐานแน่ชัด แต่คาดว่าคงมีใช้ในประเทศจีนเป็นประเทศแรกเนื่องจากเป็นชาติแรกที่ค้นพบ และนำดินปืนไปใช้งาน แต่ลักษณะระเบิดมือในช่วงแรกจะ ใช้ภาชนะขนาดเหมาะมือ น้ำหนักเบา บรรจุวัตถุติดไฟ สารระเบิด หรือสารพิษ
ระเบิดมือใน สมัยโบราณ ตัวระเบิดทำมาจากกระดาษ กระเบื้อง หรือแก้ว และบรรจุสารอันตรายต่างๆ ตั้งแต่พิษจากพืช พิษจากสัตว์ กรด ปิโตรเลียม ดินปืน หรือแม้กระทั่งวัตถุที่ได้จากพิธีทางไสยศาสตร์
อย่างไรก็ตาม ระเบิดมือในยุคเก่ายังไม่สู้จะมีประสิทธิภาพนัก ที่ปรากฏบทบาทโดดเด่นก็คือ ในช่วงสงครามโลกครั้งที่สอง ที่มีการใช้อย่างแพร่หลาย ระเบิดมือที่มีชื่อเสียงในช่วงนั้น คือ ระเบิดมิลส์ (Mills)

ระเบิดมิลส์ (Mills)เป็นระเบิดมือแบบ แตกกระจายรุ่นแรกในยุคใหม่ และใช้ในกองกำลังแนวหน้าของอังกฤษ ระเบิดมิลส์นั้นเป็นระเบิดที่มีโลหะห่อหุ้ม มีเข็มแทงชนวน และพื้นผิวเป็นร่อง นิยมเรียกกันในภาษาไทยว่า ระเบิดน้อยหน่า โดยมีพัฒนาต่อมาอีกยาวนาน

โมโลตอฟ คอกเทล (Molotov cocktail) เป็นระเบิดมือได้ คล้ายกับระเบิดขวด ที่ใช้ขวดแก้วบรรจุน้ำมันเชื้อเพลิง แล้วชุดชนวนแถบผ้าให้ระเบิด ระเบิดแบบนี้ใช้ครั้งแรกในกองทัพฟินแลนด์ ซึ่งมีอุปกรณ์ที่ไม่ก้าวหน้านัก เมื่อครั้งต่อสู้กับกองทัพโซเวียตในช่วงสงครามฤดูหนาว เมื่อค.ศ. 1939
หลักการพื้นฐานหลักการพื้นฐานของลูกระเบิดมือนั้นง่ายมาก โดยภายในลูกระเบิดมือจะ บรรจุสารระเบิด พวกดินปืน หรือ ไนโตรกลีเซอรีน ฯลฯ เป็นต้น สารระเบิดเมื่อได้รับการจุดระเบิด จากระบบจุดระเบิด สารระเบิดจะระเบิดตัวเองอย่างรุนแรง

ระเบิดมือแบบ แท่ง (Stielhandgranate)โดยมีเชื้อประทุห่อหุ้มอยู่ในกระบอกโลหะ และติดไว้กับแกนไว้สำหรับขว้าง ระเบิดชนิดนี้มีใช้ทั้งในสมัยสงครามโลกครั้งที่หนึ่ง และครั้งที่สอง ถัดมาได้มีการพัฒนาระเบิดมืออีกรุ่นหนึ่ง เรียกว่า โมโลตอฟ คอกเทล (Molotov cocktail)

โมโลตอฟ คอกเทล (Molotov cocktail) เป็นระเบิดมือได้ คล้ายกับระเบิดขวด ที่ใช้ขวดแก้วบรรจุน้ำมันเชื้อเพลิง แล้วชุดชนวนแถบผ้าให้ระเบิด ระเบิดแบบนี้ใช้ครั้งแรกในกองทัพฟินแลนด์ ซึ่งมีอุปกรณ์ที่ไม่ก้าวหน้านัก เมื่อครั้งต่อสู้กับกองทัพโซเวียตในช่วงสงครามฤดูหนาว เมื่อค.ศ. 1939
หลักการพื้นฐานหลักการพื้นฐานของลูกระเบิดมือนั้นง่ายมาก โดยภายในลูกระเบิดมือจะ บรรจุสารระเบิด พวกดินปืน หรือ ไนโตรกลีเซอรีน ฯลฯ เป็นต้น สารระเบิดเมื่อได้รับการจุดระเบิด จากระบบจุดระเบิด สารระเบิดจะระเบิดตัวเองอย่างรุนแรง
ระบบจุดระเบิดมีมากมายหลากหลายประเภท แต่แยกออกเป็น 2 ประเภทใหญ่คือ
