เกม Assassin s Creed: Brotherhood เป็นเกมแอคชั่นผจญภัยภาคใหม่ พัฒนาโดยบริษัท Ubisoft Montreal เกมภาคใหม่ได้กล่าวถึงช่วงยุคสมัยปี 1503
เมื่อกรุงโรมเป็นเมืองที่รุ่งโรจน์ นอกจากบ้านเมืองมีการปฏิรูปพัฒนาแล้ว ด้านการทหารก็มีความเข้มแข็งและอาวุธป้องกันล้ำสมัยกว่าเดิม เช่น จากเดิมที่ทหารใช้ธนู
กลายมาเป็นปืนยาวแทน เป็นต้น ผู้เล่นสามารถเดินทางและชมทัศนียภาพกับกรุงโมงที่มีอาณาเขตกว้างขวางมากกว่า เมืองฟลอเรนซ์ หัวเมืองใหญ่จากเกม
Assassin s Creed 2 ผู้เล่นยังรับบทเป็น Ezio Auditore da Firenze ต่อสู้รับมือกับ Ceasare ผู้เป็นลูกชายของตัวร้ายจากเกมภาค 2 และเป็นนายพลควบคุม
กองทัพ Papal เมื่อ Ceasare หวังล้างแค้น Ezio โดยบุกรุกเมือง Auditore และคฤหาสน์หลังโตของ Ezio และกำจัดผู้เป็นลุงของตัวละครเอก ตัวละครเอกภายใน
เกมมีพัฒนาการที่ดีตามประสบการณ์การเป็นนักลอบสังหาร ตั้งแต่การโจมตีท่าใหม่ๆและทักษะด้านปืนป่ายโลดโผน ภายในเกมมีฉากหนึ่งให้ผู้เล่นกำจัดกองทหารที่พยา
ยามใช้ปืนใหญ่ยิงถล่มหมู่ บ้าน ผู้เล่นจะต้องปืนหนีขึ้นหลังคาและกระโดดลงขี่ม้าเพื่อต่อสู้ศัตรู พร้อมกับต้องหลบหนีไปตั้งหลักเมืองอื่นแทน ศัตรูภายในเกมมีอาวุธล้ำสมัย
และรู้ทันมากขึ้น ทำให้การจัดการเพียงคนเดียวอาจเป็นไปได้ยาก ภาคนี้จึงมีเพื่อนฝูงที่มีอุดมการณ์เดียวกัน หวังโค่นล้มผู้ที่มีอำนาจที่คอยกดขี่ประชาชน ผู้เล่นจะได้พบ
กับทีมสังหารจากเมืองและทวีปต่างๆ มีอาวุธและทักษะเป็นพิเศษ สามารถเรียกใช้ทีมสังหารตามต้องการ ตามสถานการณ์อันตรายและพัฒนาตัวละครให้แข็งแกร่งมืออาชีพ
มากขึ้น ลูกเล่นรายละเอียดปลีกย่อยเกมภาคใหม่ยังเหมือนกับเกมภาคที่แล้ว ตั้งแต่การหลบซ่อนเข้าฝูงชน ว่าจ้างกลุ่มสาวๆ, โจรสมัครเล่น และเหล่าชายฉกรรจ์ ปั่นป่วน
กลุ่มทหารเพื่อหลบหนีง่ายขึ้นและสร้างจุดสนใจเพื่อเข้าไปยังบริเวณ ต้องห้าม เก็บสมบัติตามฉาก หรือซื้ออาวุธและชุดเกราะ
