eFootball PES 2021 ตัวเกมมีการอัพเดตการย้ายทีมและค่าพลังความสามารถของนักเตะอย่างติโม แวร์เนอร์, อัลฟอนโซ่ เดวีส์, ลีรอย ซาเน่, ติอาโก้ ซิลวา, ฮาคิม ซีเยค จากภาคที่แล้วให้ผู้เล่นได้สนุกสนานกันได้อย่างเต็มที่ แต่ผู้เล่นที่เพิ่งจะย้ายทีมมาคงต้องรอแพทช์อัพเดตใหม่ครั้งหน้า
เกมเพลย์ ยังคงใช้งานเอนจิ้นเดิมของ PES 2020 ทำให้เกมเพลย์เหมือนกับภาคเดิม ผู้เล่นไม่ต้องปรับตัวมากกับเกมเพลย์ สามารถเล่นและเพลิดเพลินกันได้ต่อเนื่องจากภาคที่แล้ว
เกมภาคนี้ได้รับลิขสิทธิ์อย่างเป็นทางการจาก Barcelona, Man Utd., Bayern Munich, Juventus เหมือนเดิม รวมทั้งสนามต่างๆอีกด้วย แต่สำหรับทีมและลีกอื่นๆ ก็มี Option File ให้ผู้เล่นโหลดมาลงเพื่อให้ได้อรรถรสมากขึ้น
